Effect of mevalonate analogues upon cholesterol biosynthesis

چکیده

Three analogues of mevalonic acid have been found to inhibit cholesterol biosynthesis in liver homogenates in the following order of efficacy: 3-methyl-3-hydroxy pentanoic acid > A*3methyl pentenoic acid > As-3-methyl pentenoic acid. The hydroxy acid produces half the maximum inhibition at a concentration of 5 pmoles/ml of homogenate. This corresponds to a ratio of 100 pmoles of inhibitor to 1 amole of substrate.

Feedback regulation of cholesterol biosynthesis: studies with cholestyramine.

Regulation of hepatic cholesterol biosynthesis is thought to occur at the step involving formation of mevalonate from 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA); this reaction is catalyzed by HMG-CoA reductase. Oral administration of cholestyramine, an agent that interferes with cholesterol reabsorption and is associated with a compensatory increase in hepatic cholesterol synthesis, was us...

Sites of control of hepatic cholesterol biosynthesis.

An inhibition in the conversion of mevalonate to cholesterol has been demonstrated in liver of cholesterol-fed rats by both in vitro and in vivo methods. Synthesis decreased to 30% of the control value after 1 week and 20% after 1 month on a 1% cholesterol diet. After a year, synthesis from mevalonate was almost completely inhibited. The rate of conversion of squalene to cholesterol was not con...

The effect of cholesterol on ubiquinone and tetrahymanol biosynthesis in Tetrahymena pyriformis.

The biosynthesis of ubiquinone-8 from radioactive mevalonate by cultures of Tetrahymena pyriformis is demonstrated. Under normal conditions the incorporation of this radioactive precursor into ubiquinone and the triterpenoid alcohol tetrahymanol reflects the amounts of these two compounds in the cell. Growth of T. pyriformis in the presence of cholesterol results in a complete inhibition of inc...

Influence of thyroid hormone status on mevalonate metabolism in rats.

Mevalonate, an essential intermediate in cholesterol synthesis, is metabolized either to cholesterol or, by the shunt pathway, to CO2. Previous investigations have demonstrated that the kidneys are the chief site of circulating mevalonate metabolism and that sex hormones as well as insulin markedly influence circulating mevalonate metabolism. The present study examined in rats the influence of ...

Cholesterol Biosynthesis: Mevalonate Synthesis Inhibited by Bile Salts.

Certain bile salts inhibit biosynthesis of mevalonate from acetate-2-C(14) in normal rat liver homogenates. The physiological control of cholesterol biosynthesis is discussed.
